Jawdy has it right.? Roll back the drivers to 3.2.0.0 is the key.? Win10 is the devil and by trying to "help" you with a driver update, it breaks CHIRP.?

Roll your drivers back to Prolific 3.2.0.0., if you have a Prolific chipped cable. Should work fine, then.

This is my typical Chirp workflow:
* Read current settings from rig (requires setting COM port, make, model).
* Archive "before" config for later use.
* Make necessary changes to config.
* Archive

Did you actually try to write back to the radio? While the boxes may have been grayed out, did they still show the FT-60 in the box?
