What are the ports you have defined in YAAC? You shouldn't need any Serial_TNC ports since you would use a AGWPE or KISS-over-TCP port for YAAC to talk to Direwolf.
It sounds like yu have a port misconfiguration error.

Hey guys,
Im new to aprs and am hoping to get this running for a backpacking trip in two months.
I have both direwolf and yaac able to Rx aprs packets and the gps coordinates through usb dongle. I just need to be able to Tx my position now. When i turn on YAAC the radio keys and wont unkey unless i exit. Any ideas would be greatly appreciated!
Setup goes: Yaesu vx-6r-->digirig-->usbhub-->FZ-M1 windows machine
I know an aprs capable radio would make things easier; but, im trying tk get what i currently have functional, despite the headaches haha.
