That depends on your SPROG and Operating System types. Some use FTDI, my SPROG IIv3 reports as a generic modem-class device and in the case of macOS at least, uses inbuilt Apple drivers. But

Yes, the SPROG connection *is* configured as "SPROG command station".
I don't know the interfaces her, between JMRI, SPROG driver and SPROG hardware.

Key is confirming it works from the turnout table in JMRI. If for some
reason that isn't working, the panel will clearly not work. Using the
command monitor for the connection will also help

Premis: I do not have a Tam Valley booster.
Open the turnout table, then change one turnout with the throttle and watch
if the table changes for that turnout. If it does, then click the
Close/Thrown
