I'd suspect a failure in the Prolific Cable electronics/9pin plug or the Command Station connection gone dodgy, as Marc said.
If you've covered all other bases:
- Run my Terminal command and ensured the port you have selected is correct.
- JMRI settings.
- Power Pro settings.
You don't have many other options except to follow the instructions at this link:

The JMRI settings are the same as #1 below and The connectors have never been removed or altered from the NCE station. I will check the AIU and review all again in the morning and get back to you.
