I expect the speaker you are plugging in uses + and shield ground on the cable. The TX500 has a differential audio output and the speaker must not be grounded.
Check the wiring diagrams in the handbook and you will see what I mean.

Update: The cable with headset female 3.5mm connector will not drive the speaker. If I substitute that cable with the microphone and plug the same speaker into the headset jack on the mic, then it drives the speaker OK.
Is there some difference between the two speaker/headset connections besides the mic connection being mono?
